BHV-1 induced oxidative stress contributes to mitochondrial dysfunction in MDBK cells

چکیده
The levels of cellular reactive oxygen species (ROS) and ATP as well as the mitochondrial membrane potential (MMP) in response to bovine herpesvirus 1 (BHV-1) infection of MDBK cells were measured, respectively. BHV-1 infection increased ROS production which depended on viral entry, and de novo protein expression and/or DNA replication. Vice versa, excessive ROS was required for efficient viral replication. Levels of both ATP and MMP were significantly decreased after BHV-1 infection. Interestingly, the loss of MMP was ameliorated by ROS depression. Collectively, ROS dependent mitochondrial damage and ultimately disruption of energy metabolism (ATP depletion) are a potential pathogenic mechanism for BHV-1 infection.

Background and purpose: Curcumin is a natural polyphenolic compound in turmeric (Curcuma longa). Curcumin has potent free radical scavenger and antioxidant properties that could significantly reduce oxidative damage. Oxidative stress and mitochondrial dysfunction contribute to valproate sodium induced tissue damage. This study investigated the protective effects of curcumin against valproate so...

Introduction: Neurodegenerative diseases are progressive disorders that could impair neuronal functions and structures. Oxidative stress and mitochondrial dysfunction are involved in the etiology of neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease and etc. Gemfibrozil is used as a therapeutic drug for hyperlipidemia. It has been shown that gemfibrozil is n...
متن کاملThe involvement of oxidative stress in bovine herpesvirus type 4-mediated apoptosis.
Bovine herpesvirus type 4 (BHV-4) belongs to the gamma-2-herpesviruses of the Gammaherpesvirinae subfamily. BHV-4 has a worldwide distribution and has been isolated in a variety of clinical diseases as well as from healthy cattle. In this report we demonstrate that BHV-4 induces apoptosis in MDBK cells. In the early phases of apoptosis, cells show an increase in the intracellular level of react...

Background and purpose: Resveratrol (RSV) is a naturally existing polyphenolic compound abundantly found in grapes and several plants. It has potent free radical scavenger and antioxidative properties with significant effects in reducing oxidative damage. Oxidative stress and mitochondrial dysfunction contribute to PQ induced tissue damage. In this study, the protective effect of RSV was invest...

Bovine herpesvirus 1 (BHV-1), a member of the alpha-herpesvirinae sub-family, causes significant losses to the cattle industry. BHV-1 establishes latency in trigeminal ganglionic sensory neurons, but periodically reactivates from latency. Previous studies suggested that infection with BHV-1-induced novel morphological changes in rabbit skin (RS) cells versus bovine kidney cells (MDBK). Conseque...
